SCF/c-KIT Effector Reporter Cell
| I. Background | |
受體激酶在正常細胞功能中的作用至關(guān)重要，人類有 58 種已知的受體激 酶，該家族最重要的成員之一是 c-KIT。c-KIT 也稱為 CD117，是一種由 KIT 基因編碼的 III 型受體激酶。它以干細胞生長因子（SCF）為配體，因此被稱為干細胞生長因子 受體（SCFR）。 c-Kit 的信號轉(zhuǎn)導通過其配體 SCF 蛋白的結(jié)合而被激活，通過自磷酸化引 起 c-Kit 二聚化和激活，并啟動磷酸化級聯(lián)反應(yīng)，從而調(diào)節(jié)細胞生長。c-Kit 磷酸化可觸發(fā) 多種信號轉(zhuǎn)導通路，包括 JAK/STAT、RAS/MAP 激酶通路、PI3 激酶、PLCγ通路和 SRC 通 路。SCF/c-KIT 信號轉(zhuǎn)導通路的激活通常與細胞增殖、遷移和存活有關(guān)，從而調(diào)節(jié)造血、 色素沉著和精子發(fā)生的關(guān)鍵功能。 | |

SCF/c-KIT Effector Reporter Cell 報告基因藥靶模型很好的模擬了體內(nèi) SCF/c-KIT 的信號轉(zhuǎn)導過程，原理見下圖所示。
