| I. Background | |
| T 細(xì)胞在細(xì)胞介導(dǎo)的免疫中發(fā)揮核心作用，可以介導(dǎo)長期、抗原特異性、效應(yīng)和記憶 反應(yīng)。T 細(xì)胞是許多免疫療法的靶標(biāo)，包括檢查點(diǎn)抑制劑、雙特異性 T 細(xì)胞接合器和免疫 激動(dòng)劑。此外，T 細(xì)胞本身已被研究為血液系統(tǒng)惡性腫瘤中使用嵌合抗原受體添加或自體 過繼轉(zhuǎn)移的療法。近年來，各種旨在誘導(dǎo)、增強(qiáng)和/或工程化 T 細(xì)胞反應(yīng)的策略已 成為治療癌癥和自身免疫等疾病的有希望的方法。 T 細(xì)胞活化是由 TCR 和 CD3 復(fù)合物的結(jié)合以及隨后的共刺激分子（如 CD28 受體）的 結(jié)合啟動(dòng)的。這些受體在細(xì)胞表面的共同作用導(dǎo)致細(xì)胞內(nèi)信號(hào)傳導(dǎo)事件和核轉(zhuǎn)錄因子的激 活，例如 NFAT、NFκB 和 AP-1。具體來說，TCR/CD3 復(fù)合物的參與使 PLC-γ的磷酸化， 導(dǎo)致細(xì)胞內(nèi) Ca 2+和 NFAT 的轉(zhuǎn)錄激活；而 TCR/CD3 與共刺激受體 CD28 的共同作用導(dǎo)致 ERK/JNK 和 IκB 激酶（IKK）的激活，進(jìn)而分別調(diào)節(jié) AP-1 和 NFκB 通路的轉(zhuǎn)錄激活。IL-2 啟動(dòng)子含有 NFAT、NF-kB 和 AP-1 的 DNA 結(jié)合位點(diǎn)。因此，TCR/CD3 和 CD28 的共同作 用導(dǎo)致 IL-2 的產(chǎn)生，IL-2 可被用作 T 細(xì)胞活化的檢測指標(biāo)。 | |
